Transplanting organoids with tissue-specific characteristics into immunodeficient mice such as nude mice or NSG mice enables dynamic simulation of tumor development and embryonic development processes, as well as high-throughput drug screening. The mouse transplantation also achieves in vivo validation.

Service Process

  1. Construct patient-derived tumor organoids.
  2. Perform genetic manipulations (e.g., lentiviral transfection to analyze single-gene or multi-gene combinations).
  3. Harvest the constructed organoids (approximately 2×10^5 cells in 15 μl of Matrigel) and transplant (inject) them into the corresponding site of 5-7-week-old NSG mice/nude mice.
  4. Compare and analyze tumorigenesis and the expression of genes in various pathways.

Advancements

Organoids maintain a high degree of cellular type and functional consistency with their tissue of origin. NSG mice can host a high proportion of normal or cancerous human cells. Therefore, this technology can provide patient-specific preclinical information for tumor patients and offer a convenient and indicative platform.
