PDOX Model Validates Thyroid Organoid Function: Researchers have, for the first time, established an organoid culture system that mimics the functional maturation of the human thyroid. Human embryonic thyroid organoids were transplanted under the renal capsule of NSG mice, demonstrating that the transplanted thyroid organoids possess proliferative capabilities in vivo and can secrete thyroxine (T4). This work provides a significant theoretical and model foundation for human thyroid development and prospective medical research.

PDOX Model Simulates Hepatoblastoma Initiation and Metastasis: By manipulating the Hippo-YAP signaling pathway in human fetal liver organoids, a single-gene-driven human hepatoblastoma initiation model was established. YAP1-activated fetal liver organoids were orthotopically transplanted into NSG mice, resulting in tumor formation in the liver and spontaneous lung metastases, thereby simulating the process of human hepatoblastoma initiation and metastasis.
